        BD采用*利技術,從富含胞外基質蛋白的EHS小鼠腫瘤中分離出BDMatrigel基底膜基質,其主要成分由層粘連蛋白,Ⅳ型膠原,巢蛋白,硫酸肝素糖蛋白等組成,還包含生長因子和基質金屬蛋白酶等。BDMatrigel基底膜基質在室溫條件下,聚合形成具有生物學活性的三維基質,模擬體內細胞基底膜的結構、組成、物理特性和功能,有利于體外細胞的培養(yǎng)和分化,以及對細胞形態(tài)、生化功能、遷移、侵染和基因表達的研究。

        BDMatrigel基底膜基質形成的三維培養(yǎng)基質,可促進上皮細胞、肝細胞、Sertoli細胞、黑色素瘤細胞、血管內皮細胞、甲狀腺細胞及毛囊細胞等的貼壁與分化。同時,Matrigel還能影響乳腺上皮細胞的蛋白表達,支持外周神經的新生和牛輸卵管上皮細胞的分化。高濃度的Matrigel適用于研究體內血管生成和腫瘤細胞遷移及腫瘤模型的建立等。

        BD基質膠/基質膜cornning356234特點:

        Matrigel基質會有色差變化(淡黃色到深紅色),是由于酚紅和碳酸氫鹽與CO2作用引起的,但是與5%CO2平衡后色差即會減少。凍融后,輕輕搖晃試劑瓶使Matrigel分散均勻。所有操作均需在無菌環(huán)境下進行,試劑瓶瓶蓋可用70%乙醇擦拭,并自然干燥。應使用預冷的移液器以保證Matrigel呈勻漿狀。

        細胞可在0.5mm厚度的Matrigel基質層表面生長,也可在1mm厚度的Matrigel三維基質內生長。過度稀釋的Matrigel會形成非膠質的蛋白層,可以用于細胞貼壁,但不能用于細胞的分化研究。

        注意:可將凍融后的Matrigel分裝在多個小管,所有分裝均需用預冷的凍存管,迅速冷凍并保存,避免多次凍融。

        Matrigel在22-35℃溫度環(huán)境下快速成膠,因此溶解時在4℃冰上過夜凍融(4度時會隨著溫度的上升部分成膠)。所有用品在使用前需置于冰浴,必須使用預冷的移液管、吸頭及小管操作Matrigel。成膠后的Matrigel可以在4℃24-48小時后重新呈液態(tài)。

        推薦包被與成膠方法:

        注意:為了保證Matrigel基質膜的成膠性能與穩(wěn)定性,稀釋濃度不應低于1:3,可用無血清培養(yǎng)基稀釋,Matrigel成膠后立即使用。

        薄膠成膠方法:

        1.凍融后,用預冷的移液槍頭混勻Matrigel基質成勻漿狀。

        2.將需要使用的培養(yǎng)板置于冰上,加入濃度為50μL/cm2生長面積的Matrigel基質。

        3.在37℃放置30分鐘,即可使用。

        厚膠成膠方法:

        1.凍融后,用預冷的移液槍頭混勻Matrigel基質成勻漿狀。

        2.將需要使用的培養(yǎng)板置于冰浴,將培養(yǎng)的細胞與Matrigel基質混合,用移液槍頭使其懸浮于基質中。加入濃度為150-200μL/cm2生長面積的Matrigel基質。

        3.在37℃放置30分鐘,可成膠??梢约尤爰毎囵B(yǎng)的基質,也可使細胞直接生長在膠表面。

        薄層包被方法:

        1.凍融后,用預冷的移液槍頭混勻Matrigel基質成勻漿狀。

        2.根據使用需要,采用無血清培養(yǎng)基稀釋Matrigel基質。根據實驗需要確定*包被濃度。

        3.將稀釋的Matrigel基質包被于所需的培養(yǎng)器皿中,包被量至少覆蓋整個器皿的生長表面。室溫下孵育1小時。

        4.去除未結合的Matrigel,用無血清培養(yǎng)基輕輕地沖洗。
